How the Assay Works


The RT-Direct Multiplex Real Time PCR Kit uses multiplex real-time reverse-transcription PCR technology to simultaneously detect three SARS-CoV-2 genomic targets: the RdRp gene, E gene, and N gene. A unique feature of the assay is Buffer R3, which enables direct RNA release from clinical samples without requiring a separate viral RNA extraction procedure. The buffer safely inactivates pathogenic samples while preserving and stabilizing released viral RNA, and thus reducing hands-on processing time. Guanidine thiocyanate present in Buffer R3 disrupts the viral capsid, rendering the virus non-infectious while maintaining nucleic acid integrity for amplification.

An integrated internal control monitors PCR inhibition, helping prevent false-negative results. The kit also includes a positive control template and all essential reagents required for seamless assay setup and performance validation.
