As COVID-19 continues to circulate globally, rapid and reliable molecular diagnosis remains essential for effective patient management, infection control, and public health surveillance. The emergence of viral mutations and varying clinical presentations, from asymptomatic infection to severe pneumonia, ARDS, sepsis, and death has highlighted the limitations of single-target molecular assays. Accurate multi-gene detection strategies are therefore critical to minimise false-negative results and maintain diagnostic confidence across evolving variants.
The RT-Direct Multiplex Real Time PCR Kit for COVID-19 from Genes2Me provides rapid and precise qualitative detection of SARS-CoV-2 by simultaneously targeting the RdRp, E, and N genes in a single multiplex assay. Designed with an innovative direct sample processing workflow, the kit incorporates Buffer R3, eliminating the need for a separate viral RNA extraction kit, significantly reducing turnaround time, workflow complexity, and operational costs.
The RT-Direct Multiplex Real Time PCR Kit uses multiplex real-time reverse-transcription PCR technology to simultaneously detect three SARS-CoV-2 genomic targets: the RdRp gene, E gene, and N gene. A unique feature of the assay is Buffer R3, which enables direct RNA release from clinical samples without requiring a separate viral RNA extraction procedure. The buffer safely inactivates pathogenic samples while preserving and stabilizing released viral RNA, and thus reducing hands-on processing time. Guanidine thiocyanate present in Buffer R3 disrupts the viral capsid, rendering the virus non-infectious while maintaining nucleic acid integrity for amplification.
An integrated internal control monitors PCR inhibition, helping prevent false-negative results. The kit also includes a positive control template and all essential reagents required for seamless assay setup and performance validation.
As SARS-CoV-2 continues to evolve, reliance on a single genetic target may increase the risk of missed detection due to mutations in the viral genome. Multiplex testing overcomes this challenge by simultaneously targeting the RdRp, E, and N genes, ensuring robust and reliable detection even if mutations affect one target region.
The inclusion of multiple viral targets improves analytical sensitivity and specificity while strengthening confidence in clinical reporting. The E gene supports sensitive screening, the RdRp gene provides highly specific confirmation of SARS-CoV-2, and the N gene enhances detection of low viral load samples due to its high expression levels.
Simultaneous detection of RdRp, E, and N genes ensures high analytical sensitivity and specificity while minimizing the risk of variant-related false-negative results.
Unique Buffer R3 eliminates the requirement for a separate viral RNA extraction kit, simplifying sample processing, reducing turnaround time, and lowering testing costs.
